How to prevent the formation of black spots in fetal bovine serum culture
Recently, a customer inquired about how to prevent the formation of black spots in fetal bovine serum cell culture? Our technical specialist provided a detailed explanation on how to prevent customers from generating black spots due to operational reasons. Therefore, the following points must be done well. Only then can cell culture proceed smoothly.
1、 Defrost
Remove the serum from the -20 ° C freezer and thaw it at room temperature (or soak it in tap water) for about 30 minutes to 2 hours. Alternatively, it can be thawed overnight in a 4 ° C freezer. If it is not immediately inactivated after thawing, it can be temporarily stored in a 4 ° C freezer
2、 Inactivation
Place it in a room temperature water bath and start heating (at the same time, place an empty serum bottle filled with 100ml tap water, insert a thermometer, and monitor the temperature based on this)
When the thermometer displays 56 degrees, stop heating and switch to intermittent heating, maintaining 56 degrees (± 0.5 degrees) for 30 minutes (± 3 minutes). If the temperature accidentally rises more than 56 degrees, it can still be used if it still does not exceed 60 degrees and the time is short, such as no more than 5 minutes. If it exceeds 60 degrees or the time is long, it can be discarded or used for the cultivation of some ordinary cells
Remove and allow to cool naturally to room temperature (approximately 1-3 hours). It can be packaged or frozen at -20 degrees Celsius for further storage.
3、 Sub packaging
Transfer to the sterile room and transfer the serum into 10ml centrifuge tubes on the ultra clean bench (note to gently shake the serum for several weeks and mix it well beforehand; when blowing the serum with a pipette or blow tube, be careful not to blow out bubbles (serum is very viscous and easily produces bubbles; if bubbles are produced, pass them over the flame of an alcohol lamp).
Store in a -20 degree freezer.
The entire operation takes about 4-6 hours.
