Southern blot
Experimental principle
Southern blot was created by the British southern in 1975, and it is a basic technique for studying DNA maps. Southern blot hybridization is a general method for locating specific sequences of genomic DNA. Generally, agarose gel electrophoresis is used to separate the DNA fragments digested with restriction enzymes, the DNA on the gel is denatured, and the single-stranded DNA fragments are transferred in situ to nylon membranes or other solid supports, and dried or baked or UV irradiation is fixed, and then hybridized with the labeled probe of the corresponding structure, and the color is developed by autoradiography or enzyme reaction to detect the content of specific DNA molecules.
