Key points of ELISA kit for interleukin experiments


There are various types of samples for detecting interleukins in ELISA kits, such as serum, plasma, urine, cell culture supernatant, or tissue homogenate. The preparation methods for different types of test samples are different. The processing of experimental samples can be regarded as one of the key processes in ELISA experiments, so scientific research friends must pay more attention.

1、 Concentrate
Due to the introduction of solvents during the purification process, the concentration of the tested component may decrease and may not be suitable for direct analysis. Therefore, it is necessary to remove all organic solvents. During the pre-treatment process of the reagent kit, the sample is dried under nitrogen at 60 ℃, and then the monotonic residue is dissolved in a complex solution. Concentration method: Nitrogen drying to remove impurities, compressed air drying to remove impurities.

2、 Homogenization
① Organizational samples: Cut meat and liver into small pieces, repeatedly grind them with a meat grinder, and mix evenly.
② Aquatic samples: Remove the non edible part of the sample, cut the edible part finely, and homogenize with a homogenizer; When the surface of the material is dirty, it needs to be cleaned with distilled water appropriately.
③ Eggs: Remove the shell from fresh eggs, mix the yolk and egg white thoroughly.
④ Fruits and vegetables: First wash away the sediment with water, then remove the surface moisture from the ELISA kit, and take the edible portion.

3、 ELISA kit for oscillating extraction of interleukins
Add the extraction solvent into a stoppered container containing the sample, shake it to ensure full contact between the extraction solvent and the sample inside the container to deepen into the interior of the sample tissue, and extract the component to be tested.
① Oscillatory methods: up and down oscillation, reciprocating oscillation, and hand cranked up and down oscillation on the oscillator.
② When participating in the extraction between organic solvents in tissue samples, it is necessary to oscillate while adding to avoid tissue agglomeration, which is not conducive to extraction.
