Selection and optimization of antibody dilution of Abcam

What dilution is used



The binding rate between antibody and antigen depends on the affinity constant. The affinity constant can be affected by temperature, pH and buffer composition. The formation of antibody antigen complexes can also be controlled by changing the relative concentration of antibody and antigen in solution. Since it is usually impossible to change the concentration of the antigen, the optimal working concentration (dilution) of each antibody must be determined for each application and each experimental condition.

Optimization of antibody dilution: titration experiment

However, the concentration of polyclonal antiserum antibody may be significantly different among different animals or at different time of serum collection. In these cases, or when you notice a change in staining results between batches of any antibody, we recommend that the antibody be titrated again to obtain the optimal concentration.
