Sample processing and requirements
1. Serum: Whole blood samples should be left at room temperature for 2 hours or overnight at 4 ℃, then centrifuged at 1000g for 20 minutes. The supernatant can be taken for detection, or the sample can be stored at -20 ℃ or -80 ℃, but repeated freeze-thaw should be avoided.
2. Plasma: EDTA or heparin can be used as anticoagulants. Within 30 minutes after specimen collection, centrifuge 1000g at 2-8 ° C for 20 minutes, or store the specimen at -20 ° C or -80 ° C, but avoid repeated freeze-thaw cycles.
3. Tissue homogenate: Rinse the tissue with pre cooled PBS (0.01M, pH=7.4) to remove residual blood (lysed red blood cells in the homogenate can affect measurement results), weigh and cut the tissue into small pieces. Add the shredded tissue to the corresponding volume of PBS (usually in a weight to volume ratio of 1:9, for example, a 1g tissue sample corresponds to 9mL of PBS, and the specific volume can be adjusted appropriately according to experimental needs and recorded. It is recommended to add protease inhibitors to PBS) into a glass homogenizer and grind thoroughly on ice. In order to further lyse tissue cells, the homogenate can be sonicated or subjected to repeated freeze-thaw cycles. Finally, add the homogenate to 5000 × Centrifuge for 5-10 minutes and take the supernatant for detection.
4. Cell culture supernatant or other biological specimens: Centrifuge 1000g for 20 minutes to collect the supernatant for detection, or store the specimen at -20 ℃ or -80 ℃, but avoid repeated freeze-thaw cycles.
Note: Hemolysis of the specimen can affect the final test result, so hemolytic specimens should not be subjected to this test.
