Key points to note in ELISA experiments!

1、 Important washing:
Insufficient washing can lead to differences and high background, resulting in unsatisfactory experimental results. If unbound materials remain in the microplate, it will increase background noise. If necessary, the salt concentration in the washing solution can be increased to prevent non-specific binding reactions. If the background is too high and there is suspicion of insufficient washing, you can try increasing the number of washes.

2、 Key closure:
The function of the sealing solution is to allow unrelated proteins to occupy potential binding sites in the microplate. This reduces the chance of non-specific binding of antibodies that can generate signals. If the background is too high and there is suspicion of insufficient sealing, you can try using a higher concentration of sealing solution or extending the sealing time appropriately. There are currently two main types of sealing liquids: protein and non-ionic detergents. The type of use depends on multiple factors, including the surface reagents of the microplate, adsorbed antigens, antibodies, and detection reagents. A good blocking solution should reduce non-specific binding, but it should not interact with antigens, antibodies, or detection reagents.

3、 Antibody concentration needs to be optimized:
If the reagents are slightly different, it may be necessary to optimize the amount of antibodies. Non specific antibody binding increases the background. To prevent this, do not use too many primary or secondary antibodies.

4、 The testing reagents should be in moderation:
Do not use too many testing reagents. If the concentration is too high or not diluted correctly, it can lead to a high background. The ELISA kit should not show excessive color, and if necessary, optimize when to add the termination solution. Only by ensuring that every step is done properly can perfect experimental results be presented. So every step of the ELISA experiment is important and cannot be taken lightly.
