  Whether the medium can be sterilized completely depends on the following factors:

  1. Is the air in the sterilization pot drained?

  This is the key to the proportional relationship between temperature and pressure that affects sterilization. When the same pressure is reached, if the air is not purged, that is to say, it is not pure steam sterilization, the temperature at the moment can reach the intended request. , Will seriously affect the sterilization effect.

  2, moist heat sterilization

  When    moist heat sterilization, the microorganisms are killed, and the nutrients are also damaged, especially amino acids and vitamins. For example, at 121°C for only 20 minutes, 59% of lysine, arginine and other basic amino acids were damaged, and an appropriate amount of methionine and tryptophan were also damaged. Under the effect of heat, certain nutrients may also react to each other due to heat, which constitutes a change in the quantity of the original nutrients in the medium, thus affecting its quality.

  3, foam

  The air in the foam forms a thermal insulation layer, which makes it difficult for heat to penetrate and kill the bacteria in it.

  4, particles

  The particles are small and easy to sterilize, while the large particles are difficult to sterilize. For those containing a few larger particles and coarse fibers, coarse filtration can be used to remove them.

  5, pH value

  The pH value has a great influence on the heat resistance of microorganisms. When the pH is between 6.0 and 8.0, microorganisms are least likely to die. When the pH is less than 6.0, the microorganisms die relatively easily. At this moment, H+ easily enters the cells of the microorganisms, thereby changing the physiological response of the cells and prompting their death. Therefore, the lower the pH of the medium, the shorter the time required.
